Laborszolgáltatás-03 – Sequencing 3

The present procedure is issued within the framework of the referenced DBR, in the service category of “Molecular and biotechnological procedures”, for the purpose of concluding a service contract for various sequencing services.

1. Subject matter and description of the procurement: mRNA sequencing of plant samples – 28 individual, frozen plant leaf samples, with the option for the contracting authority to order the service for up to an additional 8 samples. Expected requirements: Total RNA isolation and QC (Quality Control) • The winning bidder shall isolate total RNA from frozen plant samples provided in RNALater solution in 2 ml Eppendorf sample collection tubes, and then perform quality control on these templates. • Measurement protocols containing the results obtained during quality control shall be provided to the Contracting Authority as an electronic file. Standard mRNA-Seq execution • The winning bidder shall purify mRNA from total RNA samples with RIN≥7 quality and perform strand-specific library preparation following fragmentation. • The prepared pooled libraries shall be sequenced on an Illumina or equivalent platform with 2x150 bp PE reads, after which the winning bidder shall provide the raw data to the Contracting Authority. • The entire workflow shall be carried out in a quality-assured manner, with a guaranteed average paired-end read count of 50 million/sample. The winning bidder is obliged to provide the measurement protocols containing the results obtained during library preparation and quality control, sequencing reports, and raw data in FastQ format to the Contracting Authority. 2. Expected further minimum requirements: • The results of the quality and quantity tests related to the samples and the sequences shall be sent electronically (placed in the Illumina basespace cloud) to the contracting authority in a format suitable for further bioinformatics processing. • The contracting authority does not request bioinformatics processing. • The contracting authority requests the results by transferring FastQ files. • Data and samples are expected to be stored for at least 3 months after the examination/analysis is completed. • The winning bidder is obliged to provide the opportunity for consultation at all stages of performance. The collection, provision, and transport of samples are the responsibility of the Contracting Authority. 3. Performance deadline: Samples will be handed over once during the term of the contract. Sample handover will take place within 20 calendar days of the contract signing, at a time agreed upon in advance by the parties. The performance deadline is 40 calendar days from the receipt of the samples. The contracting authority accepts early performance. The contracting authority will inform the winning bidder in writing about the use of the option within 10 calendar days of the contract coming into effect.
